4-Amino-6-alkyloxy-2-alkylthiopyrimidine Derivatives as Novel Non-nucleoside Agonists for the Adenosine A1 Receptor
Alexandros G. SfakianakisστοΘεραπείαΠριν από 18 δευτερόλεπτα
Abstract Three 4-amino-6-alkyloxy-2-alkylthiopyrimidine derivatives (4-6) were investigated as potential non-nucleoside agonists at human adenosine receptors (ARs). When tested in competition binding experiments, these compounds exhibited low micromolar affinity (Ki values comprised between 1.2 and 1.9 μM) for the A1 AR and no appreciable affinity for the A2A and A3 ARs. Evaluation of their efficacy profiles by measurement of intracellular cAMP levels revealed that 4 and 5 behave as non-nucleoside agonists of the A 1 AR with EC50 values of 0.47 μM and 0.87 μM, respectively. No clear... περισσότερα »

Oligodendrocytes, a type of brain cell that plays a crucial role in diseases such as multiple sclerosis, are more diverse than have previously been thought, according to a new study by researchers at Karolinska Institutet in Sweden. The findings, published in the journal Science, will help increase our understanding of diseases in which these cells are affected and possibly provide clues to future treatment strategies. In multiple sclerosis and similar neurological diseases, the electrical signals in the brain propagate more slowly. The reduced speed of information flow contribute... περισσότερα »
The essential function of the Trypanosoma brucei Trl1 homolog in procyclic cells is maturation of the intron-containing tRNATyr [ARTICLE]

*Trypanosoma brucei*, the etiologic agent of sleeping sickness, encodes a single intron-containing tRNA, tRNATyr, and splicing is essential for its viability. In Archaea and Eukarya, tRNA splicing requires a series of enzymatic steps that begin with intron cleavage by a tRNA-splicing endonuclease and culminates with joining the resulting tRNA exons by a splicing tRNA ligase. Here we explored the function of TbTrl1, the *T. brucei* homolog of the yeast Trl1 tRNA ligase. We used a combination of RNA interference and molecular biology approaches to show that down-regulation of TbTrl1... περισσότερα »

The nontemplated addition of single or multiple nucleotides to RNA transcripts is an efficient means to control RNA stability and processing. Cytoplasmic RNA adenylation and the less well-known uridylation are post-transcriptional mechanisms regulating RNA maturation, activity, and degradation. Gld2 is a member of the noncanonical poly(A) polymerases, which include enzymes with varying nucleotide specificity, ranging from strictly ATP to ambiguous to exclusive UTP adding enzymes. Human Gld2 has been associated with transcript stabilizing miRNA monoadenylation and cytoplasmic mRNA ... περισσότερα »

Genomic alignment of small RNA (smRNA) sequences such as microRNAs poses considerable challenges due to their short length (~21 nucleotides [nt]) as well as the large size and complexity of plant and animal genomes. While several tools have been developed for high-throughput mapping of longer mRNA-seq reads (>30 nt), there are few that are specifically designed for mapping of smRNA reads including microRNAs. The accuracy of these mappers has not been systematically determined in the case of smRNA-seq. In addition, it is unknown whether these aligners accurately map smRNA reads con... περισσότερα »

We investigated the hypothesis, motivated in part by a coordinated computational cough network model, that second-order neurons in the nucleus tractus solitarius (NTS) act as a filter that shapes afferent input to the ventral respiratory network (VRC) during the production of cough. In vivo experiments were conducted on anesthetized spontaneously breathing cats. Cough was elicited by mechanical stimulation of the intrathoracic airways. Electromyograms of the parasternal (inspiratory) and rectus abdominis (expiratory) muscles and esophageal pressure were recorded. In vivo data reve... περισσότερα »
The action of the diaphragm on the rib cage

When the diaphragm contracts, pleural pressure falls, exerting a caudal and inward force on the entire rib cage. However, the diaphragm also exerts forces in the cranial and outward direction on the lower ribs. One of these forces, the "insertional force", is applied by the muscle at its attachments to the lower ribs. The second, the "appositional force", is due to the transmission of abdominal pressure to the lower rib cage in the zone of apposition. In the control condition at FRC, the effects of these two forces on the lower ribs are nearly equal and outweigh the effect of pleu... περισσότερα »
The Correlation Between Brain Near-Infrared Spectroscopy and Cerebral Blood Flow in Piglets with Intracranial Hypertension

Cerebral perfusion pressure (CPP) is used as a surrogate for measurement of cerebral blood flow (CBF) but its determination requires that intracranial pressure be directly measured. Near-infrared spectroscopy (NIRS) can non-invasively measure tissue oxygenation. We hypothesized that NIRS would correlate well with CBF, with cerebral metabolism of oxygen (CMRO2) and glucose and with lactate production as CPP was reduced. Seven anesthetized piglets were subjected to reductions in CPP to 60, 50, 40, 30 and 20 mmHg by infusing an artificial cerebral spinal fluid into the lateral ventric... περισσότερα »
The different effects of strength and endurance exercise training on COX-2 and mPGES-expression in mouse brain are independent of peripheral inflammation

Acute endurance exercise has been shown to modulate COX-2-expression, which is suggested to affect neuronal plasticity and learning. Here, we investigated the effect of regular strength and endurance training on cerebral COX-2-expression, inflammatory markers in the brain and circulating cytokines. Male C57BL/6N mice were assigned to either a sedentary control group (CG), an endurance training group (EG; treadmill running for 30 min/day, 5x/week, 10 weeks) or a strength training group (SG; strength training by isometric holding, same duration as EG). The standard treatment that some patients with brain cancer receive is likely to change, based on findings from two large clinical trials presented at the American Society of Clinical Oncology (ASCO) annual meeting in Chicago this week. Both trials showed that administering the chemotherapy drug temozolomide (Temodar®) in addition to radiation therapy increased how long patients lived overall and without their disease progressing. The trial investigators and other leading brain cancer researchers agreed that the results of the two trials will change the standard of care. from Ca... περισσότερα »

High-resolution colonic motility recordings in vivo compared with ex vivo recordings after colectomy, in patients with slow transit constipation
Alexandros G. SfakianakisστοSwallowing and Swallowing Disorders (Dysphagia)Πριν από 2 ώρες
Abstract Background The pathogenesis of slow transit constipation (STC) remains poorly understood, with intrinsic and extrinsic abnormalities implicated. Here, we present high-resolution colonic manometry recordings from four STC patients recorded before total colectomy, and subsequently, *ex vivo*, after excision. Methods In four female, treatment-resistant STC patients (median age 35.5 years), a fiber-optic manometry catheter (72 sensors spaced at 1 cm intervals) was placed with the aid of a colonoscope, to the mid-transverse colon. Colonic manometry was recorded 2 h before and a... περισσότερα »
Effects of hemin on heme oxygenase-1, gastric emptying, and symptoms in diabetic gastroparesis
Alexandros G. SfakianakisστοSwallowing and Swallowing Disorders (Dysphagia)Πριν από 2 ώρες
Abstract Background Therapeutic options for management of diabetic gastroparesis are limited. Failure to maintain upregulation of heme oxygenase (HO1) leads to loss of interstitial cells of Cajal and delayed gastric emptying (GE) in non-obese diabetic mice. Our hypothesis was that hemin upregulation of HO1 would restore normal GE in humans with gastroparesis. Aims To compare effects of hemin and placebo infusions on HO1 activity and protein, GE, autonomic function, and gastrointestinal symptoms in diabetic gastroparesis. Methods In a single-center, double-blind, placebo-controlled,... περισσότερα »
Effects of ghrelin receptor agonist, relamorelin, on gastric motor functions and satiation in healthy volunteers
Alexandros G. SfakianakisστοSwallowing and Swallowing Disorders (Dysphagia)Πριν από 2 ώρες
Abstract Background Synthetic human ghrelin accelerates gastric emptying, reduces gastric accommodation, and results in numerical increases in postprandial symptom scores. The ghrelin receptor agonist, relamorelin, accelerates gastric emptying in patients with diabetic gastroparesis. Aim To measure pharmacological effects of relamorelin on gastric accommodation, distal antral motility, and satiation in healthy volunteers. Methods In a placebo-controlled, double-blind, randomized study of 16 healthy volunteers, we compared effects of 30 *μ*g subcutaneous (s.c.) relamorelin to placeb... περισσότερα »

Immunobiotic Lactobacillus strains reduce small intestinal injury induced by intraepithelial lymphocytes after Toll-like receptor 3 activation
Alexandros G. SfakianakisστοΝοσολογίαΠριν από 2 ώρες
AbstractObjective Intestinal intraepithelial lymphocytes (IELs) play critical roles in disrupting epithelial homeostasis after Toll-like receptor (TLR)-3 activation with genomic rotavirus dsRNA or the synthetic dsRNA analog poly(I:C). The capacity of immunobiotic *Lactobacillus rhamnosus* CRL1505 (Lr1505) or *Lactobacillus plantarum* CRL1506 (Lp1506) to beneficially modulate IELs response after TLR3 activation was investigated in vivo using a mice model. Results Intraperitoneal administration of poly(I:C) induced inflammatory-mediated intestinal tissue damage through the increase o... περισσότερα »
