Abstract
Hematopoiesis is the process which generates all the mature blood cells from the rare pool of hematopoietic stem cell (HSCs). HSC divide by asymmetric division which provide it two distinct features of self-renewal and multi-potent differentiation capacity. Hematopoiesis is a steady state process in which mature blood cells are produced at the same rate at which they are lost, establishing a homeostasis. HSCs are regulated through their environmental niche, cytokine signalling, and the orchestrated activities of various transcription factors. The aim of this review is to summary the current knowledge of the biological functions of bioactive lipids and lipid mediators in hematopoiesis, and highlights bioactive lipid functions in megakaryocytopoiesis. Most of the existing literature involved in the studying on bioactive lipids function in HSCs showed that lipids are important regulators as mediators in various signal transduction events in hematopoiesis. Herein we reviewed the effects of PGE2 on homing, cell survival and self-renewal of HSCs, the PGE2 participate in variety of important physiological processes via G-protein coupled receptor signaling. Further, we present an overview of knowledge regarding the physiological functions of phosphatidylinositides and ceramides in cells of different hematopoietic lineages. This review also discusses current reports on lipids functions in megakaryocytes and platelets. Recent studies demonstrate that lipid signalling plays an important role in development of blood cells from HSCs. Many lipid derivatives revealed as important regulators in promoting stem cell survival and lineage commitment.
